\nP\u0159\u00edb\u011bh follistatinu za\u010dal v roce 1807 (i kdy\u017e ne\u00famysln\u011b) objevem plemene skotu v Belgii, kter\u00e9 m\u011blo nadm\u011brn\u00e9 mno\u017estv\u00ed svalov\u00e9 tk\u00e1n\u011b ve spojen\u00ed s minim\u00e1ln\u00edm mno\u017estv\u00edm t\u011blesn\u00e9ho tuku.<\/p>\n
Proto toto plemeno proch\u00e1zelo liniov\u00fdm \u0161lecht\u011bn\u00edm, dokud se nadm\u011brn\u00e1 svalov\u00e1 hmota nestala „standardn\u00edm“ znakem jejich druhu – dnes toto plemeno zn\u00e1me jako belgickou „modrou kr\u00e1vu“.<\/p>\n
A\u010dkoli jsme to v t\u00e9 dob\u011b nev\u011bd\u011bli, jednalo se o prvn\u00ed zji\u0161t\u011bn\u00fd p\u0159\u00edpad nedostatku myostatinu.<\/p>\n
Po mnoho let (ve skute\u010dnosti t\u00e9m\u011b\u0159 200) se neud\u00e1lo nic v\u00fdznamn\u00e9ho, co by se t\u00fdkalo v\u00fdvoje nebo pochopen\u00ed follistatinu – objeven\u00ed, zd\u016frazn\u011bn\u00ed \u00fa\u010dink\u016f a pochopen\u00ed molekuly myostatinu m\u011bly b\u00fdt p\u0159elomov\u00fdmi prvky, kter\u00e9 jsme pot\u0159ebovali, abychom pln\u011b pochopili, co p\u0159esn\u011b follistatin je a jak funguje.<\/p>\n
Tento objev m\u011bl trvat je\u0161t\u011b d\u00e9le – do\u0161lo k n\u011bmu a\u017e v roce 1997.<\/p>\n
A\u010dkoli jsme vid\u011bli jeho \u00fa\u010dinky v akci (nebo jejich nedostatek), jak se projevily u „modr\u00e9 kr\u00e1vy“ – prost\u011b jsme nebyli moud\u0159ej\u0161\u00ed, pokud jde o to, co p\u0159esn\u011b myostatin je.<\/p>\n
N\u00e1\u0161 objev tohoto „svalnat\u00e9ho“ plemene krav n\u00e1m p\u0159\u00edli\u0161 nepomohl, pokud jde o smysl zlov\u011bstn\u00e9 l\u00e1tky; prost\u011b jsme zjistili, \u017ee tito tvorov\u00e9 jsou „nov\u00ed“ a p\u0159edstavuj\u00ed vynikaj\u00edc\u00ed komer\u010dn\u00ed p\u0159\u00edle\u017eitost, kterou lze vyu\u017e\u00edt, ani\u017e bychom kdy pronikli pod povrch a zjistili, pro\u010d se narodili pr\u00e1v\u011b takov\u00fdm zp\u016fsobem.<\/p>\n
Teprve v roce 1987 jsme kone\u010dn\u011b „objevili“ a izolovali folistatin prost\u0159ednictv\u00edm objevu genu FST; ozna\u010dili jsme ho jako protein potla\u010duj\u00edc\u00ed FSH nebo, jak se brzy za\u010dalo zkracovat (zhruba ve stejn\u00e9m \u010dasov\u00e9m r\u00e1mci), „folistatin“ – byl objeven ve folikul\u00e1rn\u00ed tekutin\u011b prase\u010d\u00edch vaje\u010dn\u00edk\u016f.<\/p>\n
Tento objev p\u0159edch\u00e1zel objevu myostatinu o deset let a nakonec p\u0159ipravil p\u016fdu pro „situaci“ s follistatinem, v n\u00ed\u017e se dnes nach\u00e1z\u00edme, pokud jde o v\u00fdzkum, kter\u00fd prov\u00e1d\u00edme.<\/p>\n
Po v\u00fd\u0161e zm\u00edn\u011bn\u00e9m objevu myostatinu v roce 1997 se n\u00e1m poda\u0159ilo poskl\u00e1dat skl\u00e1da\u010dku, kter\u00e1 vypr\u00e1v\u011bla o interakci mezi follistatinem a myostatinem a o tom, jak tyto dv\u011b l\u00e1tky tvo\u0159\u00ed t\u00e9m\u011b\u0159 „jin a jang“ vztah, kter\u00fd podporuje hormon\u00e1ln\u00ed homeost\u00e1zu v souvislosti se svalovou hypertrofi\u00ed (r\u016fstem).<\/p>\n
Dal\u0161\u00ed pr\u016flomov\u00fd objev p\u0159i\u0161el v roce 2004, kdy Schuelke a jeho kolegov\u00e9 zdokumentovali p\u0159\u00edpad extr\u00e9mn\u00edho nedostatku myostatinu u lidsk\u00e9ho kojence.<\/p>\n
Tento p\u0159\u00edpad n\u00e1m umo\u017enil pln\u011b si uv\u011bdomit, k jak\u00fdm fyziologick\u00fdm zm\u011bn\u00e1m m\u016f\u017ee u \u010dlov\u011bka doj\u00edt, kdy\u017e je p\u0159\u00edtomnost myostatinu v syst\u00e9mu negovateln\u00e1.<\/p>\n
Krom\u011b toho jsme se (v roce 2007) setkali s dal\u0161\u00edm zv\u00ed\u0159ec\u00edm exempl\u00e1\u0159em v podob\u011b dnes ji\u017e pom\u011brn\u011b nechvaln\u011b proslul\u00e9ho psa „Wendy“.<\/p>\n
Podobn\u011b jako d\u0159\u00edve objeven\u00e1 odr\u016fda skotu se i ona narodila s t\u011b\u017ek\u00fdm deficitem myostatinu.<\/p>\n
V pr\u016fb\u011bhu t\u011bchto objev\u016f, kter\u00e9 se v p\u0159\u00edslu\u0161n\u00fdch letech uskute\u010dnily, prob\u00edhaly testy s follistatinem na zv\u00ed\u0159atech (n\u011bkter\u00e9 z nich byly v\u00fd\u0161e zm\u00edn\u011bn\u00e9 testy na my\u0161\u00edch, jejich\u017e v\u00fdsledky byly analyzov\u00e1ny v \u010d\u00e1sti „inhibice myostatinu“).<\/p>\n
Tyto testy p\u0159inesly pozoruhodn\u00e9 v\u00fdsledky, pokud jde o to, \u010deho p\u0159esn\u011b m\u016f\u017ee b\u00fdt follistatin schopen u lidsk\u00e9ho hostitele, pokud se pou\u017eije k potla\u010den\u00ed hladiny myostatinu.<\/p>\n
Teoreticky by to m\u011blo p\u0159in\u00e9st podobn\u00e9 v\u00fdsledky jako u v\u00fd\u0161e uveden\u00fdch „p\u0159\u00edrodn\u00edch“ vzork\u016f – to je to, co nakonec vedlo k pokus\u016fm na lidech, kter\u00e9 v sou\u010dasn\u00e9 dob\u011b prov\u00e1d\u00edme.<\/p>\n
